The purpose of this in vitro assay is to evaluate, via imagery, the protective effects of compounds (molecules, active ingredients, extracts) on the mitochondrial depolarization induced by a pollutant (SMR1649b urban dust)
There is evidence today that mitochondria are one of the main targets of environmental toxic substances (pollution) which can damage mitochondrial morphology and function, thus resulting in the acceleration of cell ageing and even in the development of certain pathological disorders ( e.g. metabolic syndrome).
Mitochondria play a central role in the production of ATP and metabolic cofactors (NADH, FADH2) involved in the respiratory chain. They are also involved in key processes linked to cellular differentiation and to the regulation of the cellular cycle.
In order to assess cell health, the study of membrane potential can be a discriminating indicator of the functional state of cells.
Here, the cyanine dye JC-1 (5,5′,6,6′-Tetrachloro-1,1′,3,3′-tetraethyl-imidacarbocyanine iodide) facilitates the discrimination of “energized” and “deenergized” (depolarized) mitochondria as the normally green fluorescent dye forms red fluorescent aggregates when it is concentrated in “energized” mitochondria, as a response to their higher membrane potential.
